TroFuse-005: Dr Pothuri on Sac-TMT in Advanced Endometrial Cancer

Bhavana Pothuri, MD, reviews the unmet needs in advanced or recurrent endometrial cancer, where 5-year survival is about 20%, and the design of the phase 3 TroFuse-005 study (NCT06132958). The trial compared sacituzumab tirumotecan (sac-TMT), a TROP2-directed antibody-drug conjugate, with physician’s choice of chemotherapy in 776 previously treated patients. Sac-TMT nearly doubled median progression-free survival (7.2 vs 3.9 months; HR, 0.59), improved median overall survival (19 vs 12.1 months; HR, 0.68), and nearly doubled the objective response rate. Pothuri explains why these results support sac-TMT as a new standard of care in this setting.
