Paul L. Swiecicki, MD, of the University of Michigan Rogel Cancer Center, reviews efficacy results from the fully enrolled cohort 1 of the phase 1b/2 OrigAMI-4 study, published in the Journal of Clinical Oncology in 2026. Among 102 patients with recurrent or metastatic head and neck squamous cell carcinoma evaluable for response, Dr Swiecicki reports a confirmed overall response rate (ORR) of 42% by a blinded independent reviewer, a figure he calls remarkable against historical control. He highlights the depth and durability of response, noting a complete response (CR) rate of 15% and a rate of response lasting 6 months or longer of 24%, though he notes this duration figure is still limited by follow-up time. Dr Swiecicki adds that nearly 31% of patients remained on treatment at the time of the analysis, reflecting an early readout with ongoing follow-up.
