Paul L. Swiecicki, MD, of the University of Michigan Rogel Cancer Center, shares his perspective on where subcutaneous amivantamab may fit in the treatment paradigm for previously treated, recurrent or metastatic head and neck squamous cell carcinoma (HNSCC), based on cohort 1 data from OrigAMI-4 and the agent's Breakthrough Therapy Designation. He describes potential candidates as patients who progress on chemoradiation within 6 months and then fail first-line immunotherapy, or patients who progress after first-line chemoimmunotherapy, both of whom could be considered for amivantamab in the platinum- and immune-checkpoint-inhibitor-refractory setting. Dr Swiecicki stresses that this data applies only to HPV-unrelated disease and does not extend to HPV-related or oropharyngeal HNSCC. He notes that further studies are needed to define whether EGFR bispecific antibodies have a role in these HPV-related populations in the modern treatment era.
